Cathepsin S Immunocapture Activity Assay, Fluorogenic

The Calbiochem InnoZyme™ Cathepsin S Activity Assay Kit (Cat. No. CBA050) uses a monoclonal antibody specific for human cathepsin S immobilized on a 96-well plate. Activity of the captured cathepsin S is measured at 37�C using a fluorogenic substrate, Z-Val-Val-Arg-AMC. Cleavage of the scissile amide bond within the substrate releases free AMC, resulting in an increase in fluorescence, which is measured at an excitation wavelength of 360-380 nm and an emission wavelength of 440-460 nm.

Key elements of the assay:

	Specificity: Exquisite specificity to human cathepsin S delivered through immunocapture via monoclonal antibody immobilized on a 96-well plate. Cross-reactivity with human cathepsin B, L, H, and K is less than 0.5 %.
	Sensitivity: Activity assay range 1-40 ng/ml (1-4 ng/well) allows assessment of enzyme activity in a variety of biological samples.
	Simplicity: Just add your sample, incubate for 60 minutes wash, incubate with substrate, and read fluorescence at standard AMC read-out.
	Broad Application: The assay is designed to measure enzyme activity in tissue extracts and cell lysates as well as for screening cathepsin S inhibitors.

�	Activity of cathepsin S in biological samples. The kit specifically measures cathepsin S in a variety of human tissues and cells. Cross-reactivity with other cathepsins is minimal.
