Introducing the InnoZyme™ Insulysin/IDE Immunocapture Activity Assay Kit
 
Ask A Technical ScientistInsulysin or insulin degrading enzyme (IDE) is an evolutionarily conserved, ubiquitously expressed, 110 kDa zinc metalloprotease. The enzyme is primarily cytosolic, however, a fraction of the enzyme is proximal, located on the cell surface or secreted. IDE plays an important role in diseases such as diabetes and Alzheimer's disease. In diabetes, it is the major enzyme responsible for degrading insulin. In Alzheimer's disease, IDE degrades Ab peptides associated with plaque formation.

The Calbiochem® InnoZyme™ Insulysin/IDE Immunocapture Activity Kit is a specific and sensitive assay for measuring active human, mouse, or rat IDE in cell lysates, tissue extracts, and biological fluids. The assay uses an affinity purified polyclonal antibody specific for IDE immobilized on a 96-well plate to capture the enzyme. Activity of captured IDE is measured using a FRET substrate, Mca-GGFLRKHGQ-EDDnp. Cleavage of the scissile amide bond between R and K, releases the fluorophore from the quenching molecule, Dnp, resulting in an increase in fluorescence. Fluorescence is measured at excitation 320 nm and emission 405 nm.
