APAF-1 (apoptotic peptidase activating factor) gene encodes a cytoplasmic protein that initiates apoptosis. Modulation of apoptosome function in cancer has been observed in metastatic melanomas in which APAF-1 protein was lost due to methylation-induced transcriptional silencing.

PARC (p53-associated parkin-like cytoplasmic protein), is a ubiquitin ligase that acts as a cytoplasmic anchor protein for p53. PARC directly interacts with p53 and forms a 1 MDa complex in the cytoplasm of unstressed cells. Inactivation of PARC induces nuclear localization of p53 and induces p53-dependent apoptosis.
