Whether purifying proteins from E. coli, yeast or other eukaryotic cells, the first step is always disruption of the cells and extraction of the relevant protein fraction. This step is often critical because harsh mechanical or enzymatic treatments can affect the target protein's structural integrity and activity. To address this problem, Novagen has introduced additional members of the Buster family of protein extraction reagents. These are novel detergent-based formulations that enable gentle, efficient, non-mechanical extraction of soluble proteins from bacteria, yeast, and mammalian cells. In practice, cells are harvested by centrifugation and suspended in the appropriate reagent. After a brief incubation, the sample is centrifuged and the protein extract is ready for processing.
