Notes Protein kinase C (PKC) is a family of serine- and threonine-specific protein kinases that can be activated by calcium and the second messenger diacylglycerol. PKC family members phosphorylate a wide variety of protein targets and are known to be involved in diverse cellular signaling pathways. PKC family members also serve as major receptors for phorbol esters, a class of tumor promoters. Each member of the PKC family has a specific expression profile and is believed to play distinct roles in cells. The protein encoded by this gene is one of the PKC family members. Studies both in human and mice demonstrate that this kinase is involved in B cell signaling and in the regulation of growth, apoptosis, and differentiation of a variety of cell types. Alternatively spliced transcript variants encoding the same protein have been observed. Activation is required for oxidant-induced disruption of both the microtubule cytoskeleton and permeability barrier of intestinal epithelia. Calcium-dependent involucrin expression is inversely regulated by protein kinase C (PKC)alpha and PKCdelta. Data show that activation of protein kinase D in response to oxidative stress requires two sequential signaling events, mediated by protein kinase Cdelta. Data show that protein kinase C delta (PKC delta) is located downstream of RhoA and that active RhoA and PKCdelta are both necessary for leukotriene D(4)-induced stress-fibre formation. Data suggest that phosphorylation of protein kinase C delta may act as a negative feedback mechanism to limit the overall activation of the CD98 pathway. Down-regulation of hydrogen peroxide-induced activation of this enzyme in N-acetylglucosaminyltransferase III-transfected HeLaS3 cells. EGF-mediated induction of fibronectin expression occurs at the post-transcriptional level and involves PKCdelta signaling pathway. HSP25 downregulates PKCdelta, which is a key molecule for radiation-induced ROS generation and mitochondrial-mediated caspase-dependent apoptotic events. Il-2 and polysaccharide K increased expression of protein kinase C delta. Induction of neurites by the regulatory domain of PKCD is counteracted by PKC catalytic activity and RhoA GTP-binding protein metabolism. Inhibition of PKCdelta is sufficient to lead to caspase-3-independent apoptosis in the colon cancer line, COLO 205. Interleukin-7 and transforming growth factor-beta play counter-regulatory roles in protein kinase C-delta-dependent control of fibroblast collagen synthesis in pulmonary fibrosis. Involvement of c-Src and protein kinase C delta in the inhibition of Cl(-)/OH- exchange activity in Caco-2 cells by serotonin. MARCKS-mediated neurotensin release occurs via protein kinase C-delta downstream of the Rho/ROK pathway. MMP-9 inhibition activity of resveratrol and its inhibition of JNK and PKC-delta may have a therapeutic potential in cancer. Mediation of tumor growth factor beta-1 induced collagen I expression in glomerular mesangial cells. NAG-1 expression is up-regulated by TPA in LNCaP cells through a PKC-dependent pathway involving the activation of NF-kappa B. Novel isoforms regulate human keratinocyte differentiation by activating a p38 delta mitogen-activated protein kinase cascade that targets CCAAT/enhancer-binding protein alpha. Overexpression of dominant-negative PKCdelta mutant supported an essential role in virus reactivation, yet overexpression of PKCdelta alone was not sufficient to induce lytic reactivation of KSHV. PKC delta (nPKC delta) might be predominantly involved in regulating membrane functions and survival of neutrophils associated with post ischemia reperfusion injury-induced inflammatory oxidative stress. PKC delta associates with IL6ST via Stat3 and enhances Stat3-gp130 interaction. PKC delta has a role in activating p53 transcription. PKC delta is activated in a retinoic acid-dependent manner and plays a critical role in the generation of the biological effects of retinoic acid in malignant cells. PKC delta regulates airway epithelial cell expression of NF-kappa B-dependent proinflammatory genes IL-8, GM-CSF, RANTES, and ICAM-1. PKC epsilon and delta are the main PKC isozymes involved in G alpha(q)-mediated signaling in human atria. PKC mediates VEGF-induced Akt activation in a novel signal transduction pathway through which Akt can be regulated by growth factors acting through receptor protein tyrosine kinases, and may play an essential role in VEGF-stimulated angiogenesis. PKC-delta has a role in EGF protection of tumor cells from TNF-alpha-induced apoptosis. PKC-delta plays a major role in antiestrogen resistance in breast tumor cells. PKC-delta selectively inhibits H2 histamine receptor-mediated activation of nonselective cation channels in differentiating granulocytic cells. PKC-mediated exclusion of LIMK2 from the nucleus might be a mechanism to relieve suppression of cyclin D1 expression by LIMK2. PKCdelta and PKCepsilon may offer novel targets for development of cancer preventive or therapeutic agents that selectively inactivate PKCepsilon or stimulate PKCdelta. PKCdelta functions in the activation of SAPK through a Lyn --> PKCdelta --> MEKK1 --> MKK7 --> SAPK signaling cascade in response to DNA damage. PKCdelta plays a positive role in protease-activated receptor-mediated dense granule secretion, whereas it functions as a negative regulator downstream of glycoprotein VI signaling. PKCdelta plays an important role in regulating 15-lipoxygenase expression in human monocytes and subsequently modulates the inflammatory responses mediated by 15-LO products. PKCdelta-dependent phosphorylation of caspase-3 is involved in the regulation of monocyte life span. PKD activation plays a central role in NT peptide secretion; upstream regulators of PKD include PKC-alpha and -delta and Rho/ROK. PLD1 is threonine-phosphorylated in human-airway epithelial cells by a PKCdelta dependent mechanism. PLS3 is a downstream effector of PKC-delta in the mitochondria. PMA activated the promoter activity of the 12(S)-lipoxygenase gene in cells PKCdelta but not PKCalpha, indicating that PKCdelta played the functional role in mediating the gene activation of 12(S)-lipoxygenase induced by PMA. PMA transactivates the EGFR and increases cell proliferation by activating the PKCdelta/c-Src pathway in glioblastoma cells. Phosphorylated PKCdelta, but not total PKCdelta, in dorsal root ganglion was decreased in diabetic rats. Decrease in phosphorylated PKCdelta is partly causes impaired neurite outgrowth in diabetes, and PKCdelta plays role in diabetic neuropathy. Phosphorylation of MUC1 by PKCdelta increases binding of MUC1 and beta-catenin in vitro and in vivo. Physical and functional interactions between protein tyrosine phosphatase alpha, PI 3-kinase, and PKCdelta. Protein kinase C delta is activated during engagement of the type II interferon receptor and participates in IFN-gamma signaling. Protein kinase Cdelta regulates apoptosis via activation of STAT1. REVIEW:Interaction of protein kinase C isozymes with membranes containing anionic phospholipids utilizing fluorescent phorbol esters to probe the properties of the C1 domains. Switch chimeras, containing the C1B from epsilonPKC in the context of deltaPKC (delta(epsilonC1B)) and vice versa (epsilon(deltaC1B)), were generated and tested for their translocation in response to ceramide and arachidonic acid. The cleavage and activation of PKC-delta are critical components of UV-induced apoptosis in human keratinocytes, and the inactivation of PKC-delta can promote the survival of keratinocytes exposed to UV radiation. The delta isoform of protein kinase C (PKC) functions downstream of Ca 2+ in the signaling pathway from FcepsilonRI to degranulation in RBL-2H3 mast cells. The novel varepsilon and eta and atypical zeta, but not the conventional alpha and beta and the novel delta PKCs, may be involved in the signaling pathways involved in thrombin-induced human platelet P-selectin expression. These findings indicate that signaling via PKCdelta-p38 kinase-linked cascade specifically induces expression of VCAM-1 in lung epithelium in response to TNF-alpha and that this effect is both functionally and clinically significant. This protein is responsible for constitutive and DNA damage-induced phosphorylation of rad9 protein. Thus, a deregulation in PKCdelta was associated with reduced cellular sensitivity to cisplatin.
