| Mammalian cells protect their genomic DNA against environmental and metabolic insults by a series of surveillence pathways known as cell-cycle checkpoints. These checkpoints ensure high fidelity maintenance of genomic DNA and prevent inappropriate inheritance of damaged DNA. The checkpoint programs arrest the cell cycle allowing for either DNA repair or completion of DNA synthesis. Two checkpoint serine/threonine kinases, Chk1 and Chk2, whose activity is required for the normal G2 checkpoint response to a variety of cellular stresses, play an integral role in these processes. The K-LISA™ Checkpoint Activity Kit is a rapid, sensitive ELISA-based activity assay suitable for measuring kinase activity of purified or partially purified Chk1 and Chk2 preparations, in vitro Chk1 and Chk2 inhibitor screening, and for assessing the regulation of Chk1 and Chk2 in cell signaling which may aid in discovery of potential new targets of cancer therapies. The assay utilizes an N-terminal biotinylated peptide substrate (KKKVSRSGLYRSPSMPENLNRPR) that is phosphorylated on the third serine by Chk1 and Chk2. The phosphorylated substrate is detected with a phosphoserine detection antibody, followed by an anti-IgG HRP conjugate and color development with TMB substrate. |
