Overview: Designed for fast and easy extraction of total proteins from bacteria and mammalian cells and tissues without the need for sonication or precipitation. C-PEK provides a straightforward, two-step isolation of complete proteomes in a single microcentrifuge tube. C-PEK uses optimized extraction reagents to provide improved solubilization of proteins resulting in an increased total number of spots on 2D gels. The kit includes proprietary Benzonase® Nuclease, a nonspecific nuclease leading to clear, nonviscous protein solutions and improving resolution on 2D gels. Using the C-PEK procedure, additional concentration of proteins is not necessary—extracted proteins are ready for immediate use in standard downstream proteomics applications.

Figure 1: 2DE-Gel with 200 mg Protein from E. coli JM 109. Cells from E. coli JM 109 were collected by centrifugation and sample preparation with was performed. 200 mg proteins of the whole cell extract were separated by 2DE. First dimension was done by isoelectric focusing from pH4 to pH 7 in an immobilized pH gradient gel by application of 50.000 Vh. The second dimension separation was by SDS-PAGE in a 12 % polyacrylamide gel. Proteins were visualized by silver staining.

Mammalian: The Mammalian Complete ProteoExtract Kit (C-PEK) allows sample preparation from both tissue culture cells and tissue. A representative 2D gel of proteins extracted from a tissue culture cell line is shown in figure 2 .
