 | NoShift Products • Article: ID of DNA proteins using the NoShift Transcription Assay Kit NoShift II • Back to Main NoShift Page | | | | NoShift antibody mediated detection of transcription factor activity | The original NoShift™ assay system is a microassay plate–based method for analysis of DNA binding proteins that alleviates many of the problems associated with traditional gel shift or electromobility shift assays (EMSA). The 96-well NoShift plate format is versatile and enables analysis of binding activities in less than five hours without the use of radioisotopes. A biotinylated oligonucleotide is mixed with a protein sample and the protein–DNA complex is captured on streptavidin plates. The bound transcription factor is detected with a specific antibody followed by a second antibody–HRP conjugate and chromogenic substrate.
A complete NoShift Assay is made up by combining the separately available NoShift Transcription Factor Assay Kit with a NoShift Reagent Kit. This two-component system offers flexibility and convenience.
The NoShift Transcription Factor Assay Kit, which provides one 96-strip-well streptavidin-coated plate, binding reaction buffer, antibody dilution buffer, plate wash buffer, and TMB substrate, provides enough reagents for 100 reactions. An enhanced version of the core kit is also available with the added convenience of the NucBuster™ Protein Extraction Kit for rapid extraction of nuclear proteins for use with the system. The NoShift Transcription Factor Assay Kits can also be used with user-supplied reagents for ultimate flexibility where no reagent kit is available.
The NoShift Reagent Kits include a biotinylated consensus oligonucleotide, specific and nonspecific competitor sequences, a transcription factor-specific antibody, and a secondary antibody-HRP conjugates. Specific and positive control nuclear extract are available for the c-Fos, ER-a (estrogen receptor-alpha), HIF-1a, NF-kB (p65), Sp1, STAT1, and STAT3 transcription factors. |  | |
