The Rapid Cell Proliferation Assay measures the metabolic activity of mitochondria based on the cleavage of the tetrazolium salt WST-1 to produce a soluble formazan salt. Only cells with functional mitochondria can convert the tetrazolium dye to its reduced form. Most eukaryotic cells in culture, including mammalian, plant, and yeast cell types, reduce the dye sufficiently to ensure accurate assays. For each cell type a linear relationship between the number of viable cells and absorbance is established. No washing, harvesting, or solubilization steps are required.

. 100,000 Jurkat cells per well were incubated overnight at 37°C in the presence of 0, 1, or 5 mg/ml camptothecin in RPMI medium containing 2% FBS. After the incubation 10 ml of the WST-1 labeling mixture was added and the absorbance was read after a 2.5 hour incubation at 37°C.   Typical results using NIH3T3 cells.
