Detection of human Akt1, phosphorylated on Thr308 by immunoblotting. Samples: Whole cell lysates from HEK293 cells co-expressing RasV12 and RacL61. Membranes were left untreated (lanes 1-5) or treated with lamda (l) phosphatase (lane 6). Primary antibody: PhosphoDetect™ Anti-Akt1 (pThr308) Rabbit pAb (Cat. No. 124001) (1:1000) pre-incubated without peptide (lanes 1, 5, and 6), with non-phosphorylated peptide corresponding to the immunogen (lane 2), a generic phosphothreonine-containing peptide (lane 3), or phosphopeptide immunogen (lane 4). Detection: chemiluminescence.
