Bcl-10 is an apoptotic regulatory molecule identified through its direct involvement in t(1;14)(p22;q32) of mucosa-associated lymphoid tissue (MALT) lymphomas. The human bcl-10 gene encodes a protein of 233 amino acids containing an N-terminal caspase recruitment domain (CARD) characteristic of both proapoptotic and antiapoptotic molecules. Bcl-10 is expressed ubiquitously with high expression levels in lymphoid tissues and in the developing central nervous system. Forced expression of wild type Bcl-10 in cell lines induces apoptosis and activates NF-kB. Bcl-10-initiated NF-kB can be inhibited by cotransfection of dominant-negative mutants of TRAF2, NIK, IKKa, or IkBa.
