A major lysosomal cysteine proteinase that mediates degradation of MHC class II-invariant chain complexes in human and mouse antigen presenting cells. Cathepsin S secreted by activated macrophages has the ability to process myelin basic protein and amyloid b peptide which may be relevant to the pathogenesis of Alzheimer′s disease. It can hydrolyze elastin at both acidic and neutral pH. Specific activity: ≥50 mU/mg protein. One unit is defined as the amount of enzyme that will hydrolyze Z-VVR-AMC in 10 min at 25°C, in 100 mM KPO4, 5 mM EDTA, pH 6.5 (the assay is standardized such that 0.041 nmol AMC gives a reading of 1000 RFU.
