Full-length, recombinant, human histone deacetylase 3 (HDAC3) fused at the C-terminus to a His•Tag® sequence and human SMRT (NCOR2) consisting of amino acids 395-489 fused at the N-terminus to GST and co-expressed in S. frugiperda insect cells using a baculovirus expression system. SMRT co-expression is required for full HDAC3 activity. The HDAC3-SMRT repression complex represents a novel and potentially specific therapeutic target. The purified complex is useful for the study of HDAC3 regulation and for inhibitor screening. Specific activity: ≥110 units/µg protein.
