HDAC8 phosphorylated in vivo by PKA results in decreased activity, which leads to the hyperacetylation of histones H3 and H4. HDAC8 may play a role in one of the most frequent types of acute myeloid leukemia (AML). The active deacetylase is useful in the study of HDAC8 regulation and for inhibitor screening. Specific activity: ≥10 units/µg protein. One unit is defined as the the amount of enzyme required to deacetylate 1 picomol fluorescent peptide substrate [Arg-His-Lys(Ac)-Lys(Ac)] per min at 30°C.
