Liquid. In 138 mM NaCl, 50 mM Tris-HCl, 20 mM glutathione, 10% glycerol, pH 8.0. AVOID FREEZE/THAW CYCLES. Recombinant, human SIRT3 histone deacetylase consisting of amino acids 102-399 fused at the N-terminus to GST and expressed in E. coli. SIRT3 is the only sirtuin with an apparent direct link to extended life span in humans. SIRT3 deacetylates and activates acetyl-CoA synthetase 2 within mitochondria and has been reported to localize exclusively to this organelle. The active deacetylase is useful in the study of SIRT3 regulation and for inhibitor screening. Specific activity: ≥6 units/µg protein. One unit is defined as the amount of enyme required to deacetylate 1 pmol fluorescent peptide substrate [Arg-His-Lys-Lys(Ac)] per min at 30°C. Purity: ≥90% by SDS-PAGE.
