Protein Phosphatase 2B

Liquid. In 100 mM NaCl, 50 mM Tris, 6 mM MgCl2, 5 mM DTT, 0.5 mM CaCl2, 0.025% NP-40. AVOID FREEZE/THAW CYCLES. Recombinant, human calcineurin Aa (catalytic subunit) and calcineurin B (regulatory subunit) co-expressed with yeast myristoyl-CoA:protein N-myristoyltransferase. A Ca2+/CaM-dependent protein phosphatase. The resulting highly active calcineurin is N-myristoylated on the CaN B-subunit, as is the native protein. M.W. 60,000 (A subunit) and 15,000 (B subunit). Activity: ≥50,000 units/ml. Specific activity: ≥300,000 units/mg. One unit is defined as the amount of enzyme that will release 1.0 pmol of phosphate from the RII phosphopeptide substrate (DLDVPIPGRFDRRVpSVAAE; Cat. No. 207008) in the presence of calmodulin per min at 30°C, pH 7.4. Purity: ≥95% by SDS-PAGE.
