| Nonlytic expression in insect cells plus tightly regulated E. coli expression | | | The pBiEx™ Vectors are designed to allow rapid characterization of target genes in both E. coli and insect cells, and to allow high-level transient protein expression in insect cells without the time-consuming process of creating a recombinant baculovirus. The vectors feature the same hr5 enhancer and the ie1 (immediate early) promoter to direct expression in insect cells as the pIEx™ Vectors, and also allow for expression in E. coli by the tightly controlled T7lac promoter. For target protein expression, pBiEx recombinant plasmids can be transferred into a DE3 host that allows induction with IPTG. Expression can also be induced in hosts such as NovaBlue by infecting with lCE6, a phage that constitutively expresses T7 RNA polymerase. Three vectors offer a variety of N-terminal and optional C-terminal fusion tags as well as choices for no fusion tags.

InsectDirect™ System- pBiEx™-3 DNA The pBiEx™ vectors are designed for cloning and high-level expression of proteins in both bacteria and in transiently transfected Spodoptera-derived insect cells. In E. coli, target gene transcription is regulated by the tightly controlled T7lac promoter, and a ribosome binding site (rbs) directs translation initiation. In insect cells, transcription is driven by the AcNPV-derived hr5 enhancer and IE1 immediate early promoter.
pBiEx-3 contains C-terminal S•Tag and His•Tag coding sequences. For sequencing or PCR of recombinants in pBiEx-3, refer to the table below to choose the appropriate primers.
