Generating expression plasmids starting from the Gateway® Nova pET-62-DEST™ vector requires recombination with pENTR clones that carry their own ribosome binding site (RBS) and translation initiation site. If the pENTR clone lacks a stop codon and is appropriately designed for a C-terminal fusion, the target gene in the resulting pEXPR clone will be fused to consecutive C-terminal His•Tag® and Strep•Tag® II coding sequences. The consecutive purification tags enable the use of consecutive "gentle" purification procedures. |
