Aggrecanase 1 Liquid. In 150 mM NaCl, 50 mM Tris-HCl, 5 mM CaCl2, 0.05% BRIJ® 35 Detergent, pH 7.5. AVOID FREEZE/THAW CYCLES. Recombinant, human, truncated ADAM10 (amino acids 213-579) fused at the C-terminus to a His•Tag® sequence and expressed in S. frugiperda insect cells using a baculovirus expression. ADAMTS4 (a disintegrin and metalloproteinase with thrombospondin motif-4) belongs to a novel family of extracellular proteinases that hydrolyze aggrecan, the major proteoglycan of articular cartilage. Useful for the study of extracellular proteoglycan degradation, screening for inhibitors of proteoglycan hydrolysis, and as a standard in enzymatic and immunochemical assays. Specific activity: ≥1.5 U/mg protein. One unit is defined as the amount of enzyme necessary to hydrolyze 1.0 nmol of recombinant aggrecan interglobular domain (Cat. No. PF114) per min at 37°C, pH 7.5. Purity: ≥90% by SDS-PAGE. Ref.: Tang, B.L., et al. 2001. Int. J. Biochem. 33, 33. Tortorella, M.D., et al. 2000. J. Biol. Chem. 275, 18566.
