 Detection of human IRS-1, phosphorylated at Tyr612, by immunoblotting. Samples: Whole cell lysate from CHO-T cells transfected with human IRS-1, left untreated (lane 1) or treated with insulin (50 nM for 5 min) (lanes 2-5). Primary antibody: PhosphoDetect™ Anti-IRS1 (pTyr612) Rabbit pAb (Cat. No. PS1010) (0.5 µg/ml) pre-incubated without peptide (lanes 1,2), with a generic phosphotyrosine-containing peptide (lane 3), non-phosphopeptide corresponding to the immunogen (lane 4), or phosphopeptide immunogen (lane 5). Detection: chemiluminescence.
