Detection of eIF2Be, phosphorylated at ser539, by immunoblotting. Samples: Recombinant eIF2Be treated with lambda phosphatase (lane 1) or whole cell lysates from controll cells, spiked with GSK-3b (lanes 2-5). Primary antibody: PhosphoDetect™ Anti-eIF2Be (pSer539) Rabbit pAb (Cat. No. PS1017) (1:1000) preincubated without peptide (lanes 1,2), with non-phosphopeptide corresponding to the immunogen (lane 3), a generic phosphoserine-containing peptide (lane 4), or immunogen phosphopeptide (lane 5). Detection: chemiluminescence.
