Detection of human RhoA, phosphorylated at Ser188, by immunoblotting. Samples: RhoA GST fusion protein left unphosphorylated (lanes 1,3) or phosphorylated in vitro by PKA (lanes 2,4,5,6). Primary antibodies: Anti-RhoA (detecting total Rho; lanes 1,2) and PhosphoDetect™ Anti-RhoA (pSer188) (Cat. No. PS1089) (1:1000) preincubated without blocking peptide (lanes 3,4), preincubated with phosphoRhoA (pSer188) blocking peptide (lane 5), and preincubated with nonspecific phosphoserine containing blocking peptide (lane 6). Detection: chemiluminescence.
