Detection of human FAK, phosphorylated on Tyr ⁵⁷⁷, by immunoblotting. Samples: Whole cell lysate from vanadate treated chick embryo fibroblasts plated on fibronectin. left untransfected (lane 1) or transfected with human FAK (lanes 2-6). Primary antibody: PhosphoDetect™ Anti-FAK (pTyr⁵⁷⁷) Rabbit pAb (Cat. No. ST1011) (1:1000) pre-incubated without peptide (lane 1, 3), or with phosphopeptide immunogen (lane 2), non-phosphopeptide corresponding to the immunogen (lane 4), phosphopeptide derived from the corresponding region of Pyk2 (lane 5), and a generic phosphotyrosine-containing peptide (lane 6). Detection: chemiluminescence.
